    Uit de deal van vorig jaar met Gilead over GLPG3121 :
    The deal will give Gilead an option over Gapalagos’ GLPG3121, a JAK1/TYK2 inhibitor aimed at inflammation which recently engtered Phase I. The TYK2 space has seen growing interest from investors, not least because of the potential in a number of autoimmune conditions and the fact they have become an area of interest for antitrust regulators in the pending Bristol-Myers Squibb-Celgene deal. The UK’s Sareum (SAR) has a preclinical TYK2/JAK1 inhibitor, SDC-1801, which is one of the few unpartnered assets in this particular class.

    Gilead maakt gebruik van deze optie, maar wat dit financieel voor GLPG betekent is de vraag.

    In jaarverslag 2019 (en nadien ook bevestigd in conference call) stond dus dat GLPG 3121 was stopgezet:
    We recently stopped a Phase 1 study with GLPG3121, a JAK1/TYK2 inhibitor targeting inflammation, due to an undesirable PK profile.
    Het was ook niet meer opgenomen in de glossary van het jaarverslag, wat meestal betekent dat de compound inderdaad in de vuilbak is verdwenen. Maar GLPG3121 dook dus terug op in de pipeline en nu dus ook in het jaarverslag 2020:
    Our JAK1/TYK2 program with GLPG3121
    We discovered GLPG3121 as a selective JAK1/TYK2 inhibitor. This asset is currently undergoing Phase 1 studies and preclinical data point to potential application of GLPG3121 in inflammatory diseases.

    “This asset is currently undergoing Phase 1 studies”
    Dit is toch een beetje vreemd. “Studies” in het meervoud, maar er is maar 1 studie met GL PG3121 in clinicaltrials en dat is dus de “Completed” study, waarvan gezegd was dat ze stopgezet was:

    GLPG3121 is undergoing preclinical and Phase 1 studies.

    The aim of the study is to examine the safety and tolerability of GLPG3121-modified release formulation when given to healthy male subjects once as a single dose or multiple times over a period of 14 days in fasting condition or after a standard breakfast.

    The study will evaluate how the body absorbs and breaks down GLPG3121, and how GLPG3121 and the major breakdown product of GLPG3121 are eliminated from the body. In addition, the study will investigate the effect of food (high-fat) after a single oral dose of GLPG3121 as modified release tablet.

    KOL feedback on market share potential for Galapagos assets.

    The consensus view from KOL conversations was that being late to market in a crowded space is not likely to result in a substantial level of market share, unless these assets demonstrate a substantially differentiated efficacy and/or safety profile to other TYK2 inhibitors.
