   Geen vlees of vis, zo lijkt het op het eerste gezicht. Waarschijnlijk wel goed genoeg om mee door te gaan? Maar vooral omdat de lat bij ccRCC laag ligt. De vraag is of ARWR een drug heeft of een franchise. De poel lijkt me wat klein om daar al te stellige uitspraken over te doen.

   Het werkt in ieder geval beter dan dat spul van ProQR kunnen we stellen .....als je appels met peren mag vergelijken. Theoretisch kun je natuurlijk ook niet verwachten dat de tumor erg snel krimpt. Als de aangroei gestopt kan worden heb je al heel wat. Op de beurs is (gezien de opleving van het sentiment) oo niet veel begrip voor de uitkomsten. Niet hematologische kankers zijn taaie rakkers. AFMD weet er ook alles van.

   Er is in elk geval forse knock-down in de tumor. Als je dat ook hebt in early stage tumoren en in andere soorten tumoren, dan ben je spekkoper.
   Het lukt ARWR in elk geval als een van de weinigen of wellicht de eerste om RNAi de tumor in te brengen. Zelfs al is de huidige kandidaat niet sterk genoeg, dan verbeter je die. Je krijgt hem in elk geval op zijn bestemming.
   RNAi in de lever is ook ooit zo begonnen. Dit zal ons niet meteen boven de 100 brengen. Maar heeft wel het potentieel om ons ooit van bv. 300 naar 400 te brengen.

   Per 31 december was slechts 12,1% van ADAP in handen van institutions. Ik zou zeggen dat als het dubbele van de huidige beurskoers wordt geboden de deal wel rond is. Er is kennelijk nagenoeg geen belangstelling voor dit bedrijf ondanks Dieter en Dirk hun oproepen om het aandeel vooral te kopen. Toch raar zou ik zeggen.

   NB, ik had graag vanavond gekocht maar ik ga niet lenen.
   Einde maand lukt het wel en dan kan ik nog eten,leven en vooral hopen.

    - they calculate the % with UK ordinary shares which they should divide by six to get $ADAP ADS shares.
    So simply multiply the NASDAQ percent total with 6. (the count & value for individual inst owners is correct)

The data presented provide initial proof of target engagement based on reductions in hypoxia inducible factor-2 alpha (HIF2a) expression, as well as an acceptable safety profile in response to escalating doses of ARO-HIF2.     Responders in Cohort 1 (225 mg, n=3), Cohort 2 (525 mg, n=4), and Cohort 3 (1050 mg, n=2) achieved mean reductions of HIF2a protein of -45%, -57%, and -80%, respectively
    Tumoral expression of HIF2a messenger RNA (mRNA) was assessed by quantitative polymerase chain reaction (qPCR)

    Safety
    
    ARO-HIF2 was generally well-tolerated in patients. Anemia and hypoxia, frequently reported on-target adverse events (AEs) with small molecule HIF2a inhibitors, were reported in 12% of patients
    Five serious AEs in 5 patients were reported by investigators as possibly drug related, including myocarditis (in a patient with a history of TKI induced cardiomyopathy), demyelinating neuropathy (in a patient with autoimmune sequelae due to checkpoint inhibitors), chronic inflammatory demyelinating polyradiculoneuropathy (in a patient with distant history of checkpoint inhibitor use), hypoxia (in a patient with a pulmonary infiltrate), and acute hypoxemic respiratory failure (in a patient with progressive lung metastatic disease)

    1, 2, 3, respectively.
    
    Nu is het dit:
    
    qPCR: Among patients with evaluable biopsy, 9/9 showed reductions in HIF2a mRNA
    Cohort 1, Cohort 2, and Cohort 3 achieved mean reductions of HIF2a mRNA of -38%, -28%, and -44%, respectively
    IHC: Responders in Cohort 1 (225 mg, n=3), Cohort 2 (525 mg, n=4), and Cohort 3 (1050 mg, n=2) achieved mean reductions of HIF2a protein of -45%, -57%, and -80%, respectively.
    
    De cut-off datum is dus gewijzigd en de cohort 3 is groter geworden (van 1 naar 2) én laat betere data zien (zie vetgedrukt). Dat laatste is erg bemoedigend in mijn ogen. :-) Hoewel de aantallen nog veel te klein zijn voor echte conclusies (slag om de arm).

    Dat is wel iets om in de gaten te houden natuurlijk. Merck deed het overigens niet beter:
    
    here is mrk's safety record: The most common all-cause adverse events (=20%) were anemia (86.9%), fatigue (57.4%), headache (36.1%), dizziness (31.1%) and nausea (24.6%). Grade 3 all-cause adverse events included fatigue (4.9%), anemia (3.3%), dyspnea (1.6%) and weight increase (1.6%).

    This is big in so many ways....we have proof of concept for cancer, and as ca has said time and again, this opens the door for a cancer franchise.
    
    Further, we have proof of concept outside the liver, the holy grail of rnai.
    
    We delivered the payload outside the liver, and it had the desired effect. proud to be an investor today.
    
    A partnership for this drug, or this franchise is most likely in the works right now.
    
    Buiten de minpunten hierboven lijkt deze PB toch wat meer positieve signalen vrij te geven.
    Laten we dat maar hopen.

    Ja, als blijkt dat 1050 mg de optimale dosis is, en Chris weet dat ws allang aangezien de data van 1 dec is (2,5 mnd geleden), dan zijn de gesprekken achter de schermen reeds gevoerd of ze zijn er druk doende mee. Komt er binnen een week of drie geen partnerschap, dan was het allemaal slechts een toevalstreffer. Maar daar reken ik eerlijk gezegd niet op.
    
    Ik ben voorzichtig optimistisch, optimistischer dan een week geleden. :-)

    Dit zou een zakelijke stilte kunnen ingehouden hebben waartoe hij eventueel verplicht was op zijn Conference call.
    De toekomst zal het zeker wel gaan uitwijzen.
