    Lung diseases like IPF are associated with an increased chitinase activity, which strongly correlates with disease stage, progression and prognosis. Chitinases (predominantly CHIT1) are involved in macrophage activation. Inhibition of chitinase activity translates into a potential therapeutic benefit, as shown in a range of preclinical models. GLPG4716 has shown robust anti-fibrotic activity in multiple animal models, when compared with the standard of care.
    
    GLPG4716 (formerly OATD-01 inlicensed from OncoArendi) is a novel, small molecule CHIT1/AMCase inhibitor targeting a key pathway in tissue remodeling. It has shown compelling translational data, a favorable profile in animal studies at expected therapeutic doses and it has successfully completed Phase 1 studies in healthy volunteers. Galapagos aims to bring GLPG4716 to a Phase 2 clinical trial for the treatment of IPF and possibly other diseases with a fibrotic component.

    Tijdens de Galapagos-conferentie gaf het bij de bespreking van OATD-01 vooral de verdere
    ontwikkeling van het project bij IPF aan. Is het realistisch dat ze geïnteresseerd raken in de
    ontwikkeling van het project op het gebied van sarcoïdose?
    
    Marcin Szumowski: Galapagos zal zich zeker focussen op IPF, omdat het hun strategische
    ontwikkelingsrichting is. Het contract voorziet ook in andere richtingen die verband houden met
    longziekten met fibrose, waaronder sarcoïdose. Het plan is dat naast de hoofdindicatie, en de
    grootste fase 2-studie in IPF, in de loop van de tijd andere kleinere studies zullen worden uitgevoerd bij patiënten met andere ILD.
    
    Hoe geavanceerd is uw chitinaseplatform in vergelijking met andere biotechbedrijven en wat
    hoopt u op moleculen van de tweede generatie?
    
    Marcin Szumowski: We kunnen alleen verwijzen naar wat zich in het publieke domein bevindt. Er zijn geen verleende octrooien of gepubliceerde octrooiaanvragen in de wereld die dubbele of selectieve chitinaseremmers beschermen. Zeker, er is wat onderzoek gaande op dit gebied, en na de transactie met Galapagos zullen deze sterk geïntensiveerd worden. Als chitinase-remmers een therapeutisch effect laten zien in fase 2-onderzoeken, zullen zeker veel biotechnologie- en farmaceutische bedrijven in de wereld dit gebied betreden. Ik denk echter dat we op dit moment zo'n 5 jaar voordeel hebben in chitinase ten opzichte van anderen en samen met de Galapagos willen we dit voordeel behouden.
    
    Wanneer kunnen we belangrijke vorderingen verwachten met betrekking tot de tweede generatie moleculen van het chitinaseplatform?
    
    Rafal Kaminski: We hebben een aantal moleculen van de tweede generatie van het
    chitinaseplatform. Ze bevinden zich allemaal in een veel vroeger ontwikkelingsstadium dan OATD-01 en ik denk dat ze over 1,5-2 jaar de klinische onderzoeksfase kunnen ingaan. Sommigen van hen vallen onder de Galapagos-licentieovereenkomst en de stemming voor wanneer en in welke
    indicaties zullen worden ontwikkeld, behoort toe aan de Galapagos. Deze projecten zijn voor ons
    interessant vanuit wetenschappelijk oogpunt, maar het is niet zonder betekenis dat ze vanuit
    financieel en investeringsoogpunt nieuwe financieringsbronnen kunnen openen in de vorm van
    financiering van onderzoekswerkzaamheden bij OncoArendi en, na aanvang van klinische proeven,
    betalingen voor opeenvolgende verbindingen die zijn bereikt door mijlpalen.

    Nu blijkt dat Galapagos ook de global rights heeft voor FIB992 voor Lung, Liver and Kidney fibrosis.

    Ik had zopas een technieker in huis .Hij lijdt aan erge systemische sclerose.Hij deed mee aan de GPLG 1690 (ziritax) trial gedurende 2 jaar , zijn ziekte was onder controle, hij was zeer blij met de resultaten en had geen specifieke bijwerkingen.Op bepaald moment moest hij van de prof in Gent de dosis afbouwen over 14 dagen.
    Hij is echt depressief dat hij ziritax niet meer kan nemen.
    Misschien komt hij in aanmerking voor de chitinase remmer GPLG 4716 ? de fase 2 wordt er een voor IPF maar er kunnen ook andere indicaties toegevoegd worden.Lezen over de ziekte is nog iets totaal anders dan iemand voor u hebben die moedeloos wordt doordat zijn experimenteel goed werkend geneesmiddel afgenomen wordt.

    e-therapeutics plc (LON:ETX) has reached two key milestones in its collaboration with Galapagos NV, triggering two payments to the company.
    
    The two operational and success-based milestones were defined in a collaboration agreement signed in June 2020.
    
    Galapagos, a commercial-stage company focused on the discovery and development of small molecule medicines, has been using e-therapeautics’ proprietary computational platform which allows ‘in silico’ testing of computational platform.
    
    The platform has been used to successfully identified hit compounds against a specific biological process of interest to Galapagos involved in idiopathic pulmonary fibrosis and potentially in other fibrotic indications, e-therapeutics said.
    
    Also, experimental testing of the computationally predicted compound set was carried out in several relevant assays and yielded a hit rate orders of magnitude higher than industry standards for early phenotypic drug discovery, it added.
    
    "e-therapeutics' ability to model human disease processes effectively in silico addresses critical limitations of the drug discovery process,” said Ali Mortazavi, e-therapeutics chief executive.
    
    “Our deeper understanding of underlying disease biology and ability to test millions of possible therapeutic interventions computationally ahead of experimental validation have the potential to mitigate late-stage, costly failures in drug development.
    
    “The progress made to date in our collaboration with Galapagos to discover novel therapeutics in IPF and fibrosis is yet another validation of our technology platform, which can be applied to any therapeutic area."
