   Goat milk could one day be used as the latest line of defence against nerve gas poisoning, a new study suggests.
   
   Nerve gas and some agricultural products contain toxic organophosphates (OP), which upon entering humans overstimulate and poison the nervous system.
   
   Although humans make an enzyme called butyrylcholinesterase (huBChE) that can bind to the OP and prevent their effects, the amount produced is not enough to counteract the doses likely to be encountered from chemical warfare agents.
   
   The enzyme can be purified from humans, but this produces limited yields.
   
   Canadian and US researchers studied whether animals with appropriate genes inserted into them (known as transgenic animals) could excrete the enzyme in its milk.
   
   When they inserted the gene for the human form of the enzyme into the embryos of goats the resulting females were all healthy and produced large quantities of huBCHE in their milk.
   
   The enzyme was then purified and injected into guinea pigs, where it remained active in the bloodstream.
   
   Writing in the Proceedings of the National Academy of Sciences journal, the researchers argue their findings show that it is feasible to create enough huBChE from transgenic goats for potential treatment of organophosphate exposure.
   
   "The use of transgenic goats… will provide a reliable source of this enzyme for prophylaxis or treatment of humans exposed to OP compounds used as agricultural or chemical warfare agents," they conclude.

   An enzyme known to act as a powerful antidote against organophosporous compounds has been expressed in the milk of transgenic goats, report researchers in Canada and the US. Organophosphorus compounds are recognised as potential chemical weapons agents.
   
   The human enzyme butyrylcholinesterases (BChE) has long been investigated as a powerful antidote against organophosphorous compounds that might be used as chemical weapons. The goat-milk approach produces larger quantities of enzyme than can be obtained from human serum.
   
   Transgenic goats-milk wards of chemical weaponry
   
   Yue-Jin Huang and colleagues at PharmAthene Canada, Montreal, first used guinea pigs to check whether they could get mammals to produce human BChE in their milk. After a successful trial, they stepped up to an economically realistic model. Goats are often used in the production of recombinant proteins, such as antibodies, because they are cheap to maintain and breed fast.
   
   Huang's team could produce up to five grams of the antidote per litre of goats' milk. But the recombinant enzyme purified from this source did not show the characteristic tetramer assembly of the protein isolated from human serum, and did not carry the full set of sugar attachments (glycosylation pattern) necessary.
   
   The researchers then attached polyethylene glycol to the protein, which resulted in a product that was comparable in its biological activity and half-life to the original human protein.
   
   Patrick Masson, who investigates the connection between cholinesterases and organophosphate toxicity at CRSSA (Centre de Recherche des Services de Santé des Armées) at Grenoble, France, welcomed the new development. 'This is an important piece of work which considerably improves the prophylaxis and the treatment of organophosphate poisoning,' he told Chemistry World. 'It is not only of interest for the military, security staff, and medical personnel, but also for the general population who could become victims of attacks using such compounds,' he said, recalling events on the Tokyo subway in 1995, when the organophosphate sarin was responsible for 12 deaths and 50 serious injuries.
   
   'Injection of a dose of 200mg enzyme protects humans against up to five times the LD50 [dose required to kill half the members of a tested population] of the most potent nerve agents, such as soman and VX, with no side effects,' said Masson. VX nerve gas (O-ethyl-S-[2(diisopropylamino)ethyl] methylphosphonothiolate) and soman (O-pinacolyl methylphosphonofluoridate) are used solely as chemical weapons.

    Koers zit nog steeds in de bandbreedte van mijn nieuwe systeem(1.06-1.02)dus klopt allemaal nog precies.Het systeem geeft ook aan dat hij langdurig onder de 1.02 moet zijn alvorens hij doorzakt naar de 0.97 dus kan ook nog weken duren.

    Feit is wel, dat hier nog een lange weg dient te gaan, maar eens die afgelegd is, is het hier wel bingo.

The combined royalty and manufacturing transfer payment payable by Leo to GTC on commercial sales of ATryn is in the mid-to-high teens as a percentage of end-user sales, and this figure is net of GTC’s production costs. If we assume to be conservative that a 2.5% royalty is payable by GTC to third parties for IP licenses (the size of these payments has not been disclosed), then the net (pre-tax) income to GTC comes out to about 15% of Leo’s end-user sales.
    
    3.) For ATryn used by Leo in clinical trials, Leo will reimburse GTC for its cost of production up to an undisclosed cap. (Unlike commercial sales, there is no built-in profit margin for GTC on Leo’s use of ATryn for clinical trials.)
    
    4.) Leo will fund 100% of the phase-2 development in the DIC indication. When this program enters phase-3, GTC will have the option—but not the obligation—to fund half of the clinical-trial expenses. If GTC exercises this option, it will have access to all of Leo’s phase-3 data for use in the U.S. and Japan—territories not covered by the Leo partnership; if GTC declines to exercise the option, it will have access to only the phase-2 data. (GTC’s decision to exercise or not exercise this option will presumably depend on how important the European data will be to the FDA and the Japanese regulators.)

    Met een beetje geluk, zitten hier toch een paar blockbusters bij, maar goed we zien of ondergaan het wel. En als dat management nu nogeens ten gepaste tijde hun snavels opendoen, komt het misschien 'ooit' nogwel eens goed!?
