    Ik vond het wel grappig om te horen dat filgotinib inderdaad een woordspeling voor feelgoodtinib werd genoemd door Onno bij het BRT interview, iets wat ik destijds op het forum suggereerde om het voortaan feelgoodtinib te noemen.

    Bijzonder interessante presentatie die zal gegeven worden op het CF congres, volgende week in Phoenix, AZ

    Ik kijk nu al met veel ongeduld uit naar de fase 2 studie met GLPG 1837 die binnenkort zal starten. Deze studie zegt veel over het potentieel van GLPG 1837 in vergelijking met Kalydeco.

    Premature termination codons (PTCs) in CFTR result in nonfunctional CFTR protein and are the proximate cause of ~11% of CF causing alleles. Previous studies have shown that the readthrough (RT) of PTCs and CFTR protein expression and function was restored by use of aminoglycosides (AGs; gentamicin, G418) and synthetic AGs (NB124). The CFTR potentiator ivacaftor (VX-770) was successfully used in restoring CFTR function in G551D and other related mutations. We have also shown that ivacaftor can augment CFTR function following RT by potentiating rescued CFTR channels. However, ivacaftor may have deleterious effects on CFTR stability, thus the evaluation of alternative potentiators is warranted. In this study, we tested if novel CFTR potentiators alone and in combination with translational RT could improve CFTR activity in comparison to ivacaftor.

    Stably transduced Fischer Rat Thyroid (FRT) G542X or R1162X epithelial cells when fully confluent, were pretreated with G418, NB124 (250?µg/mL) and transepithelial conductance (Gt) was recorded before and after CFTR stimulation by addition of forskolin (FSK, 100 nM or 10?µM) followed by investigational potentiators (GP-1, GP-2, GP-4; 10?µM) and VX-770 (10?µM). Similar studies were also carried out in primary HBE (human bronchial epithelial, F508del/G542X) cells for short circuit current (Isc) analysis. Furthermore, potentiators were also tested on FRT CFTR missense mutations with amino acid (AA) substitutions at the 542 position (Arg, Trp, Cys) that were created to study the functional consequences of AAs inserted during PTC suppression.

    GP-2 (0.67?±?0.07 Gt) and VX-770 (0.42?±?0.05) showed CFTR potentiator activity in wild-type (WT) CFTR, with GP-2 exceeding the effect of VX-770 by 63%. Different doses of FSK pre-stimulation (100 nM, 10?µM) did not significantly affect potentiator activity. In FRT G542X cells, ivacaftor (0.1?± 0.003), GP-2 (0.15?±?0.003), and GP-4 (0.1?± 0.005) augmented CFTR function compared to vehicle control (P<0.05), but only when RT was induced by NB124 or G418 pretreatment (P<0.01). In both FRT G542X and R1162X, GP-2 CFTR activity was significantly greater than VX-770 (P<0.05). Potentiators with or without RT agents had no effect on FRT cells without CFTR, confirming specificity. In FRT G542 AA mutants, GP-2 was active and directly proportional to the degree of CFTR function with FSK stimulation alone and also more efficacious compared to VX-770. In primary HBE G542X cells, GP-2 induced Isc (3.7?± 0.6?µA/cm2) was significantly greater in NB124 pre-treated cells compared to VX-770 (2.3?± 0.5?µA/cm2; P<0.05); total stimulated current (GP-2?+?FSK) approached 22.8% of WT CFTR.

    In conclusion these results indicate that the novel potentiator GP-2 is highly efficacious towards enhancing CFTR function following translational RT of PTCs and is superior to ivacaftor in this regard, even during chronic administration. Combination therapy (potentiators?+?RT agent) may be a useful approach to augment nonsense mutation CFTR function rescued by translational RT.

    Jep, ik verkoop iig niets totdat die presentatie geweest is. Zelfs nog geen PB over uitgebracht. Meen me te herinneren dat IR had gezegd dat die abstract niet alle data geeft die gepresenteerd zal worden. Ben benieuwd, hoewel over het algemeen dit soort presentaties weinig invloed heeft op de koers. CF is een hele interessante markt, en zou een hele goede verbreding zijn van de pipeline.

    In terms of preferred dosing, Galapagos believes that efficacy must be maximised and plans to start with a bi-daily dosing of 100mg during the first 12 weeks, followed by a once-daily 200mg dose, demonstrating its firm belief in the 200mg dosing regimen.

    NIEUWE DEAL

    Because Galapagos has sufficient cash and has demonstrated its strength in big pharma deal-making, we expect an upfront inlicensing fee of $ 250m to be a minimum (with AbbVie, it was agreed to pay $ 200m for RA and an additional $ 50m for Crohn’s). However the reality may be higher, especially because Galapagos has no financial stress and can again organize a bidding process amongst the interested parties as the product is totally unencumbered.

    OPPORTUNITY AND THREAT
    Galapagos is now a take-over target. With the most valuable asset fully-owned and uncertainty expected in the coming months, any party interested in filgotinib will calculate the cost of acquiring this cash-rich company.
    The other early clinical projects may also suddenly have a value for a potential acquirer as those interested in JAK-inflammation are probably also interested in the early clinical stage projects (GLPG1690 in IPF and GLPG1205 targeting GPR84 with the phase 2a ulcerative colitis readout expected in 1H16). For some, a take-over would be considered as a threat, but for many others a bid accompanied by a good premium would be considered as an opportunity.

    De VERGELIJKING VAN DE RESULTATEN VAN ABT-494 en FILGOTINIB

    The best way to compare the ACR results of ABT494 with those of filgotinib is via the placebo correction. The table below lists these corrections for the three highest doses of ABT494, the four highest doses of filgotinib, and the working doses for humira, Xeljanz and baracitinib.

    From this data it is clear that filgotinib 2x100mg systematically has better numerical values than all 4 JAK inhibitors for both ACR20, 50 and 70. Only Humira does better for ACR20, though this TNF-blocker underperforms at ACR70. AbbVie indicated that it is proceeding with the once-daily dosing while Galapagos said it wants to start with the twice-daily 100mg dosing, so we believe the results of ABT494 24mg QD should be compared to filgotinib at 100mg BD.

    Goed bezig NielsjeB, bedankt.
    Dit zou best eens voor zeer positieve aandacht kunnen zorgen.
    2 weken geleden daalde Vertex toch aardig wat procenten alleen al op het gerucht dat Galapagos hele goede resultaten bekend ging maken voor het CF programma. Als je omrekent hoeveel hoeveel Vertex in waarde daalde dan geeft dat wel te kennen hoe waardevol dit programma is.
    Vd Stolpe hamert er al een jaar op dat dit programma zeer succesvol kan worden en als maar een klein deel van de waarde van Vertex in de koers van Galapagos tot uiting kan komen, zal dit al een flinke koersstijging kunnen opleveren.

    Update 11:15 uur: Vragen Galapagos
    Zoals al een aantal keer eerder vermeldt ben ik bij de BNR uitzending met de CEO van Galapagos en blog ik vanaf daar. Geen garantie dat het lukt, maar ik zal zeker proberen één of meerdere vragen te stellen.
    Zelf ben ik niet zo thuis in de biotech of Galapagos en ik hoor altijd dat ik er niets van begrijp, dus... Daarnaast zijn wij een site van beleggers voor beleggers en daarom vragen we u om uw diepgaande vragen in de comments te zetten. Mogelijk krijg ik de kans om er nog één of meerdere te stellen stellen. Laat dat dan hele goede vragen zijn.
